Succinate metabolism: a promising therapeutic target for inflammation, ischemia/reperfusion injury and cancer

نویسندگان

چکیده

Succinate serves as an essential circulating metabolite within the tricarboxylic acid (TCA) cycle and functions a substrate for succinate dehydrogenase (SDH), thereby contributing to energy production in fundamental mitochondrial metabolic pathways. Aberrant changes concentrations have been associated with pathological states, including chronic inflammation, ischemia/reperfusion (IR) injury, cancer, resulting from exaggerated response of specific immune cells, rendering it central area investigation. Recent studies elucidated pivotal involvement SDH immunity beyond processes, particularly context cancer. Current scientific endeavors are concentrated on comprehending functional repercussions modifications, specifically pertaining SDH, cells operating hypoxic milieu. The efficacy targeting alterations manipulate cell hypoxia-related diseases demonstrated. Consequently, comprehensive understanding succinate’s role metabolism regulation is crucial effectively therapeutic interventions influence progression diseases. This review provides succinct overview latest advancements emerging processes. Furthermore, explores succinate, intermediary TCA cycle, IR particular emphasis mechanisms underlying accumulation. critically assesses potential modulating accumulation milieu means combat various It targets by focusing disorders.

Succinate metabolism: a new therapeutic target for myocardial reperfusion injury.

Myocardial ischaemia/reperfusion (IR) injury is a major cause of death worldwide and remains a disease for which current clinical therapies are strikingly deficient. While the production of mitochondrial reactive oxygen species (ROS) is a critical driver of tissue damage upon reperfusion, the precise mechanisms underlying ROS production have remained elusive. More recently, it has been demonstr...

HMGB1: A Promising Therapeutic Target for Prostate Cancer

High mobility group box 1 (HMGB1) was originally discovered as a chromatin-binding protein several decades ago. It is now increasingly evident that HMGB1 plays a major role in several disease conditions such as atherosclerosis, diabetes, arthritis, sepsis, and cancer. It is intriguing how deregulation of HMGB1 can result in a myriad of disease conditions. Interestingly, HMGB1 is involved in cel...

Aurora-A Kinase as a Promising Therapeutic Target in Cancer

Mammalian Aurora family of serine/threonine kinases are master regulators of mitotic progression and are frequently overexpressed in human cancers. Among the three members of the Aurora kinase family (Aurora-A, -B, and -C), Aurora-A and Aurora-B are expressed at detectable levels in somatic cells undergoing mitotic cell division. Aberrant Aurora-A kinase activity has been implicated in oncogeni...
